Expected use

The ACA ELISA is designed for use in the detection of anticardiolipin antibodies in human serum or plasma. ACA can be used as indicators of antiphospholipid antibody syndrome (including thrombosis formation), spontaneous abortion, thrombocytopenia, and diagnosis of CNS lesions.

The IgG positive type for ACA may indicate that patients with cerebral thrombosis; It also results in a miscarriage. The incidence of thrombocytopenia in ACA-positive patients is significantly higher than negative and is related to the degree of platelet reduction, and ACA-positive is significantly associated with SLE patients with central nervous system (CNS) thrombosis.

Summary and explanation

The purified cardiolipin antigen is coated onto the surface of the microwells. Patient serum is added to the wells and ACA, if present, binds to the antigen during incubation. After washing the wells to remove unbound sample, the HRP-conjugated human antibody is added. Unbound conjugate is removed by a subsequent wash step.

A TMB reagent solution is then added to the microwells. The catalytic reaction of the enzyme conjugate stops at a specific time. The intensity of the color generated is proportional to the amount of ACA in the sample.
