Mechanism of Action

RNA STAT-60 employs the classic guanidinium thiocyanate–phenol single-step extraction method:

• Cell & Tissue Lysis: Rapidly denatures proteins and solubilizes cell membranes.
• RNase Inactivation: Instantly destroys endonucleases and RNases to preserve total RNA integrity.
• Phase Separation: Addition of chloroform generates a biphasic mixture where pure RNA remains in the upper aqueous phase while DNA and proteins partition to the interphase/organic layer.
• RNA Precipitation: Reagents allow rapid alcohol precipitation with isopropanol to yield high-purity, intact RNA pellets.

Typical Extraction Workflow

1. Homogenize tissue or lysate sample directly in RNA STAT-60.
2. Add chloroform, mix thoroughly, and centrifuge for phase separation.
3. Carefully transfer the upper clear aqueous phase to a clean tube.
4. Precipitate total RNA by adding isopropanol.
5. Wash the RNA pellet with 75% ethanol and air-dry.
6. Dissolve pure RNA pellet in RNase-free water or DEPC-treated water.
