Summary and explanation
The purified ds-DNA antigen is coated onto the surface of the microwells. Patient serum is added to the wells and the ds-DNA antibody, if present, binds to the antigen during incubation. After washing the wells to remove unbound sample, the HRP-conjugated human antibody is added. Unbound conjugate is removed by a subsequent wash step.
A TMB reagent solution is then added to the microwells. The catalytic reaction of the enzyme conjugate stops at a special time. The intensity of the color generated is proportional to the amount of ds-DNA antibody in the sample.
